    Overview

    PE-22-28 is a synthetic pentapeptide derived from spadin, a naturally occurring TREK-1 blocker. It has enhanced stability and a longer duration of action than spadin. Water-soluble but parenteral or intranasal administration is required due to poor oral bioavailability.

    Mechanism of Action

    PE-22-28 selectively blocks TREK-1 potassium channels, enhancing neuronal depolarization, synaptic transmission, and firing. It promotes serotonin signaling and upregulates BDNF, supporting neuroplasticity and neurogenesis, and modulates the HPA axis to reduce stress-induced damage.
