P21 was designed to isolate and enhance the neurogenic and neuroprotective activities of CNTF. In 3xTg-AD mice, chronic P021 treatment restored cognitive function, increased neurogenesis and synaptic markers, and reduced Aβ and tau.
P21 (P021, Peptide 6) is a synthetic tetrapeptide mimetic of ciliary neurotrophic factor (CNTF). It is proposed to act primarily in the CNS, particularly the dentate gyrus, mimicking CNTF effects while avoiding the adverse effects of full-length CNTF.
In mouse studies, 60 nM per gram of feed has been used. Human dosing is not established.
Safety in humans has not been assessed. Rodent studies up to 18 months show no weight loss, tumors, or pain signs seen with full-length CNTF.
